Tsunade has the health of a Snorlax, and the speed of a Raichu. Her physical strength and defense are maxed, but her special defense is not looking so hot. Zetsu, being the grass type he is gets 1/2 damage from fighting and electric. I think Zetsu wins this with sunny day and solar beam combo.

If Sakura ( before her power up ) was able to oneshot Zetsu's clone with a simple punch, then it is safe to assume Tsunade can oneshot the original Zetsu.

Zetsu isn't absorbing her chakra. To use the spores he needs prep time. And even if you give him preptime, Tsunade is a chakra beast. She could still kill Zetsu before he could deplete her chakra.
