She doesn't need to run from him. If he uses Water Dome again after she escapes she simply escapes again except this time she goes on top on it and stands on it like regular water.

The thing you fail to understand is that even if Tsunade escapes the Dome, it will still be there surrounding Kisame.

But in this particular case Water Dome won't fail. Actually, it's the exact opposite. It will force Tsunade to retreat, using her chakra to summon Katsuyu and make her split into little fodders. And all the little Katsuyu and Tsunade will have their chakra drained by Water Dome simultaneously.
