I have said this before....and I'll say it again. Gai doesn't even need the gates to beat Tsunade. Since they are from the same village, he obviously knows about her regeneration. All he needs to do is blitz her and take her head off with a kunai.(Im sure a kage level shinobi like Gai has access to one) Game,set and match.

Tsunade has not shown that she possesses durability above the average ninja, nor has she shown regeneration on the level of being able to recreate lost limbs, as seen when she could not regenerate after Madara split her in two.
