I always see people bringing up the whole rock/paper/scissors, snake/slug/frog argument for the reason why Tsunade > Orochimaru. But this is clearly false.
For starters, I'd just like to say we've NEVER seen Orochimaru at full power. I'm willing to say at least half. He specializes in Ninjutsu, and his arms and ninjutsu have been taken from him for basically the whole series.
That being said, armless Oro not only fought against Tsunade, but was also fighting JIRAIYA, another legendary sannin, on equal terms at the same time. Armless.
Imagine if Oro had his ninjutsu? Neither of them would have stood a chance in Hell. The techniques Orochimaru has developed number in the hundreds, and I doubt he'd waste his time creating weak techniques.
Another argument for Tsunade is that she is too durable for Orochimaru. But guess what? Orochimaru is JUST as durable. If she manages to get a pounding on him, it'll just be Oral Rebirth, change a strategy, and try again.
Let's not forget Orochimaru has one of the greatest solo feats of the series leagues ahead of Tsunade; fighting a fully enraged tailed beast (Again, WITHOUT ninjutsu, his whole specialty). Tsunade can't hold a candle to that.
Fact of the matter is, no matter what Tsunade throws out Orochimaru, he can counter it and best her. Tsunade is good, but she's not Orochimaru good.
So: Orochimaru > Jiraiya>=Tsunade.
