the answer is there can't be a direct biological connection between tsunade and naruto.
Minato was trained by Jiraiya.
Which makes minato too old to be tsunade's son, since jiraiya and tsunade are close in age, and minato was old enough to graduate from the ninja academy at that time.

This is the ages so you don't get confused.
Tsunade got pregnant when she was 15, give birth to Minato at age 16. Also explains sudden tit enlargement.
When she was about 21~23, Nawaki died. Minato that time is around age 5~7.
She went to ame with her team and became a sanin. Wen back to konoha to have affair with Dan. Jiraiya stayed for couple of years(3?) to train the yahiko and co. before he returned to konoha to be a jonin sensei of Minato's team.
Minato's official graduation age is 10. Tsunade this time is a bout 26 years old.
Minato died very young, 20~25 while Naruto is few hours old. Tsunade this time is about 36~41.
Naruto was 12 when he found Tsunade so that means Tsunade is around 36~41 + 12 = .48~53.

No She is not stop writhing what has ben disproved over and over fail thread.I read a couple of threads where they theorize that Tsunade has a direct blood relation with Naruto and although most of their "proof" are kinda weak, I still can't get this feeling off me that somehow they really are related.
When I first saw Tsunade, I actually thought maybe she's Naruto real mother because of their similarities but that was proven false when Kushina was introduced.
Still I can't help but think she is related with him through Minato.
Here's what I think:
First I'd like to establish how Minato got his features from Tsunade even not considering the father's side. It could be possible if Tsunade's mother/father is a Yamanaka who has blue eyes which is also the reason why Tsunade have bloned hair although one of her parents is a Senju-Uzumaki brown/read hair and it also supports the next point I'm going to make.
Sarutobi Hiruzen: Minatos' FATHER?
There are 2 instances that this could happen first is by accident.
As her sensei, Hiruzen was the one who is responsible for teaching Tsunade the skills needed for seduction mission and what better way to do it than have *** with her right? lol. He got so excited he forgot to cum outside so he got her pregnant. There is no canon evidence that a contraceptive jutsu exists.
Second reason is RAPE. Yup, ****ing monkey bastard raped Tsunade. Hiruzen got dumped by his girlfriend that time and so he got stone drunk. Tsunade found him almost passed out and out of the goodness of her heart, brought him to his apartment. But no good deed goes unpunished so the heartbroken Hiruzen who is also sexually frustrated for not having any for the last couple weeks let his lust consume him and raped poor Tsunade and got her pregnant.
Regardless of which of the above event happened, Tsunade still got pregnant. Her Yamanaka parent convinced her not to get abortion and when she gave birth to the baby they give him to the orphanage and her mother/father erased her memories of being raped, getting pregnant and giving birth. This also explains why Tsunade miraculously developed large breasts and why she has animosity/uncaring attitude to her sensi even after his death because she subconsciously hate him for impregnating her.
So Minato is born with feature of Yamanaka blond hair, blue eyes and some features of Hiruzen spikey hair. This also explains why he is super awesome and have super fast reaction time (showed when he battled A in a manga flashback). His yamanaka genes give him superior mind abilities that he don't need raiton jutsu to stimulate his nerves to have fast reaction time.
If all of this is true, then Naruto is not only related to the Yondaime but to all the previous Kages. Uzumaki Naruto is born to be hokage :scorps:

As for the bust size, puberty explains the bust size increase.
Finally, there would have been some sort of confirmation of this if tsunade had a kid.
I think jiraiya may have even specifically noted that tsunade's never had a kid when he talks to tsunade just before going to find Pain.
