Ok, so there's been a page in the manga which has been of interest to me:
Notice the word "mitotic" and "regeneration". If the translation is correct, the word "mitotic" stems from mitosis. Mitosis is:
"The process by which a eukaryotic cell separates the chromosomes in its cell nucleus into two identical sets, in two separate nuclei." - Wikipedia.
It's cell separation into two from a single cell. So if this turns out to be true, Tsunade in Byakugou mode can regenerate from a single cell. It's also follows the same idea as her summon Katsuyu, who can divide and multiply herself itself into smaller slugs.
This also explains the rules, some of them:
- Rule No. 1: Never gonna give you up from healing
- Rule no. 2: Never gonna enter a battle
- Rule no. 3: Always be the last to die
So about Rule no. 2, medics entering a battle puts the medic's life in danger and by extension, everyone else that she covers, but since Byakugou grants battle immortality, she can enter into battle.
About Rule no. 3, she can now practically "die" because if this whole mitosis idea plays out like it would, she'll "come back to life" from regeneration.
I'm totally not certain why she'd abandon Rule no. 1 unless Byakugou gives her linked AoE healing to all party members or stuff.
