So, in the bottom right panel Kakashi collected blood from Kakuzu in a vial, because of Shikamaru's plan in which he would trick Hidan into cursing Kakuzu. But...why would Kakashi do that? He thought Kakuzu was already dead since he just pierced his heart with a Rakiriki, and didn't know Kakuzu had five hearts...

If memory serves, the lightning blade to the back was done "in the moment". The vial of blood was planned ahead of time by Shikamaru, so one thing led to the other.

He didn't have to know that he had 5 hearts. The vial of blood was basically so Hidan would take care of Kakazu for them.

None of you answered my question. Shikamaru's plan was to have Kakashi collect some of Kakuzu's blood, so later in the fight the team could trick Hidan into cursing Kakuzu, and therefore kill Kakuzu. However, Kakashi thought he had already killed Kakuzu with his lightning blade, so why did he think it was necessary to get a vial of blood from Kakuzu if he thought Kakuzu was already dead?
