Yep, despite the fact that manga explicitly states that Tobirama was the one who killed Izana i still have doubts regarding this matter.
Why? Let's break it down into 3 points:
1) Itachi is never wrong.
Remember the conversation between Itachi and Sasuke? Itachi told Sasuke that Madara stole Izana's eyes and we even got the panel where Madara plucks the eyes out of Izuna's skull:
So why we would trust Itachi? Because he once told the truth regarding Kyubi incident while Tobi lied saying that was just natural disaster which was proven wrong. So it means Itachi never lied regarding madara.
2) Tobi clenching.
When Tobi had conversation with Sasuke about Itachi he clenched his arms for some reason. Many people thought that he was upset but I doubt that Obito would be upset about Izuna:
As you can see when Tobi spoke about Izuna he clenched his arm and Sasuke noticed that. He was suspicious of Tobi's words.
3) Madara's words.
When Madara pierced Sasuke with his sword he implied that he sacrificed his brother for power. His brother was Izuna
All i'm saying that Madara killed Izuna himself but blamed Senju and Tobirama for that.
