As we can see the ancestor Dōjutsu is straight from the rinnegan.
Mixed with the hatred of The juubi it caused the sharingan. ( Uchiha clan.)

3 Tomoe sharingan is powerlevels under the Ancestor Dōjutsu.
Thats why you have to kill a loved 1
and steal your brothers eyes the power of hatred cause great powers.

As kabuto stated after you obtain EMS the natural evolution of EMS is > Rinnegan.

We know the Sage was a godlike person with immense powers.
Rinnegan was his Dōjutsu. wich means he was able to surpress the hatred of the juubi. (The tomoes)

The elder son had a more twisted Dōjutsu wich means the juubi's hatred is within him.
With his hatred towards his brother he created Sharingan.

So The sharingan evolves to EMS wich give you a way to go back to Rinnegan with ''senju'' DNA

( In my opinion i think its just hashirama DNA because it looks like he came from the ''Ootsutsuki'' clan wich make sense because its the Ootsutsuki clan who stole the fruit.

Thats why Sasuke vs Naruto was destinated.

Juubi's hatred (Sasuke sharingan) Vs Naruto (Senju/Ootsutsuki) Bloodrelatives. )

EMS => Ancestor Dōjutsu.
Rinnegan > Ancestor Dōjutsu.
Rinnegan > EMS.

The Elder son's eye seems to be a eye that was sealed up by a Uzumaki technique similar to how Naruto has kyuubi sealed up with a uzumaki seal, probably when the elder son gave into the Juubi's anger it turned into tomoes
