yes, the manga says that it seals anything it pierces
I think of it more of a chakra absorber in that case. It wouldn't "seal" it for good. The person could get Susano'o right back. But it would take it away. And if it stabs something that has a soul, it seals that.

I did a bit of background reading and on NarutoPedia it says : "The Sword of Totsuka is an ethereal weapon, with an enchanted blade capable of sealing anything it pierces". So I understand that if it pierces another person's Susanno it will seal it but not forever -> The user of the Susanno probs will be able to create Susanno again.... *sighs* U_U
Susanoo is created by the users chakra, so if somehow someway susanoo can be sealed than the user could just manifest another one.
Susano is made of chakra therefore i doubt that sword can pierce it because its not solid. It will most likely go through it. Best thing to do is to aim for the user instead xd
Even if it does seal it, the user should be able to activate it again because its a ninjutsu
