[Theory] The third hole in Obito's mask and Madara's next power up.

Could it have been intended to hold the nine tomoe rinnegan? Of course neither he nor Madara have unlocked it, but Madara's Rinnegan was unnaturally made by fusing his and Hashirama's cells. It takes on the form of Hagoromo's because it is his chakra, but it looks like the nine tomoe is the true final eye since it is the juubi's eye as well as Kaguya's, while Hagoromo and Madara's lack the tomoes and thus look incomplete.

Perhaps this means Madara evolved his eyes incorrectly and required Hagoromo's yin chakra like sasuke rather than Hashirama's to make a complete Rinnegan. He fusing his mangekyou with his brother's may have spoiled the evolution as well since EMS doesn't appear to parallel the initial sharingan evolution, thus Obito planned to use the juubi to fuse his regular mangekyou with the rinnegan to make the complete nine tomoe version, and that may have been what his third eye hole was for; but it didn't work because he was unable to capture all the bijuus, did not absorb the shinju and had Hashirama's chakra like Madara rather than Hagoromo's. With this in mind, now that Madara has absorbed the Shinju's true form, or Kaguya, (whomever was speaking to him) and has all the bijuus, he may be able to manifest the nine tomoe himself now soon.

The 9 tomoe eye is Sharingan, actually.

The Rinnegan was a result of mutation caused by Hagoromo's genetics and Juubi's Chakra. That is why the Rinnegan was one of the requirements to unlock the 9 tomoe Sharingan.​
