They really had no excuse to mess with the Uchiha the way they did. The Uchiha actually turned their backs on Madara, the person who could have saved them, for the sake of following Hashirama's ideals and it ended in their extermination. The Uchiha had shown a clear sign of loyalty and yet the Senju brushed it off for the sake of doing such. Quite odd that the clan cursed with hatred had the balls to put it all aside for the future of the village.
I think that the Senju were envious of the Uchiha from the beginning. They tried mastering as many ninja arts as possible to make up for their lack of everything and still couldn't topple the Uchiha until Hashirama, an anomaly, was born. I think the Senju feared and wanted the Sharingan for themselves, but after realizing the horrible price the Uchiha's paid for the power they just found a reason to hate them even more.
The Senju were a really petty clan, I guess it's what happens when you're descended from the inferior younger brother.
