Madara's Rinnegan ( or Obito's/Nagato's ) for the matter, are probably the OLDEST functioning eyes in the Narutoverse. If we follow this timeline

97 Years BSS - Births: Hashirama Senju (P), Mito Uzumaki (P) and Madara Uchiha (P).
96 Years BSS - Births: Tobirama Senju (P), Izuna Uchiha (P), Kinkaku (P), Mū (P) and the Second Mizukage (P).
Madara was born approximately 100 years ago, with Izuna being born 99 years prior.
These eyes were the first, alongside Madara's, to awaken the Mangekyou.

The wielder of these eyes, Izuna, was said to be equal to Madara in every aspect of ninja ability. When Madara went blind due to overusing his MS, Izuna chose to sacrifice his eyes and sight so that Madara can continue to lead their clan.

Hashirama Senju, the '' God of Shinobi '', whom he fought dozens of times but was never able to beat.
There are also many and very plausible theories that Madara was the reason Tobirama died, since many argued that Tobirama, Hiruzen, Danzo, Shisui's dad and the others in Tobirama's team would be able to beat 20 ninjas.

Afterwards, these eyes eventually evolved into the Rinnegan, which Madara used to summon the Gedo Mazou and used it to build Zetsu.
The eyes would eventually be passed to Nagato Uzumaki.

Nagato's list of kills is rather impressive.
First, the two shinobi that killed his parents.

Secondly, the one who tried to kill Yahiko.
Thirdly, Hanzo/Danzo's fodder/ANBUs, through usage of the Gedo Mazo.

Several of his 6PoP were kills he made himself.

Afterwards, he killed Hanzo and everyone that was affiliated with him.

He then fought and brutally murdered Jiraiya.

Afterwards, he journeyed to Konoha, where he ( more or less ) killed a throng of fodder shinobis, caused kakashi's death, killed Shizune, and almost killed Hinata.
He then used the very same eyes to resurrect every dead person in Konoha, dying in the process.
Ironically, the most murderous eyes in the Narutoverse claimed the life of their owner.

Next on the line, Obito Uchiha murdered Konan and took Nagato's eyes. He implanted the Rinnegan into his Jins, making them his Six Paths.

Tobi is the current owner of the Rinnegan, though I assume it -will- be passed on.
In their remarkable existence ( near 100 years ), these eyes have killed more than any other jutsu:
- Hundreds/thousands of Senju Shinobi ( via Izuna and Madara )
- Defeated Mu and Onoki
- ( Supposedly ) killed Tobirama. Perhaps even Hashirama.
- Started the Eye of the Moon Plan.
- Killed hundreds/thousands of fodder shinobi from Ame and Konoha.
- Killed Hanzo.
- Killed Jiraiya.
- Killed Kakashi, Shizune.
- Almost killed Hinata.
- Last but not least, claimed the life of their user, Nagato.
