Yes but she has her healing abilities... So heal abilities + body of the Sage...

Tsunade's healing ability is limited though. It was stated in the manga when she was fighting Oro that each cell can onlyregenerate limited amount of times. So all Gai has to do is keep attacking her. He doesn't even need Gates until later in the fight. He has her beat in speed and stamina (base mode anyway). So if she has to keep healing she will run out of that ability sooner or later. And as for one punch kill only if she super punches every time she attacks and again that uses chakra too so if she misses she is pwned. She isn't just gonna go for KO punches every single punch.
