Tsunade has more physical strength than Aye, also her medical ninjutsu, her yin seal & her summon gives her an edge over Aye.

Aye is pretty faster than Tsunade and speed counts as a great weapon in a battle. Aye can hit Tsunade easily but Tsunade has her medical ninjutsu yin seal to heal her quickly. She just needs 1 or 2 hits to bring Aye down & this she can do with her physical strength, yin seal and her summon (it will be high difficulty win for Tsunade).
