• Tobirama has seen a Kagutsuchi before.
• Somebody in his generation used both Amaterasu and Kagutsuchi.
• They kinda sucked at using Kagutsuchi lol

Madara and Izuna were the only known Mangekyou Sharingan users during Tobirama's generation. So its safe to assume Izuna or Madara used either of these techniques

Also, just because he said it's hard for an experienced duo, doesn't mean they sucked. The fact that others managed to do it successfully implies there were some skilled MS users in the past.

Plus, I doubt Madara and Izuna were the only ones in Tobirama's generation to have MS. The first, yes. but not the only ones. Itachi said many over the years sought Mangekyou, and the Uchiha Bros. discovery of how to awaken it probably led to others following suit.

I think it was said in the DB that Izuna possessed Amaterasu and Kagutsuchi but I can't be certain.

I would assume it was a feat that Izuna's eyes had since we haven't seen Madara use Enton/Amateratsu even once.

They're not the only one's though since Kaguya has seen it too. Or perhaps she also has it so she commented on how Sasuke has it too.

Well if you are counting that filler then yes there were others. and perhaps there may have been other MS users between Madara/Izuna's time and Obito/Itachi/Shisui's time.

But Cannon wise Madara and Izuna were the 1st and only two to awaken the MS. Which is why Madara was chosen to be the leader of the Uchiha and I assume Izuna as his 2nd. None were really mentioned after that until Obito/Itachi/Shisui

No I didn't include the filler. Itachi had used Izamani and that means that someone else had to have developed it. And is was made specifically for MS users. Mainly, the ones who used Izanagi.
