Background: Tsunade was born as the grandaughter of Hashirama (The first Hokage) and was the grandniece of Tobirama (The second Hokage), for this reason people tended to call her "Princess". She was on Hiruzens (The future third Hokage) team with Orochimaru and Jiraya as her teammates. During the Second Shinobi War, Tsunade and her team fought Hanzo and lived, giving them the title of "The Legendary Sannin". Throughout the war Tsunade frequently counteracted Lady Chiyo's poisoning attempts, forcing Chiyo to respect Tsunade. The Leaf won the war due to Tsunades Medical and Fighting prowess. On her younger brothers twelfth birthday, Tsunade gave him a crystal necklace, in hopes of having him achieve his dream of becoming a Hokage in the future. The next day, her brother died as a casualty of the war, in which the necklace was given back to Tsunade. This gave her the idea to incorporate medical ninjas to every team while on the battlefield. But the idea was rejected because they were in a time of war and they couldn't train ninjas to become medical ninjas due to the amount of time and resources it took. A man named Dan brought it up to the Hokage again because he strongly believed Tsunade had the right idea in the fact that people should focus on saving other lives. Due to this, Tsunade grew close to Dan because they had many similarities in the way that they think and their ideals. Shortly after, the two fell in love and Tsunade then gave Dan the crystal necklace she had given her younger brother in hopes that Dans dreams would also come true. Not too long after that, Dan died a graphic death on the battlefield and once again, the necklace was returned to Tsunade. Due to this, Tsunade became afraid of blood and it made her unfit to fight in anymore battles, also people started to see the necklace as a curse and anyone who wears it dies shortly after. Since the two people she loved most in her life died, she lost faith in the title of Hokage and thought that only a fool would take on that position. So, she left the village with Dans niece, Shizune, who becomes her apprentice and one of the few people Tsunade fully trusts.

Ninjutsu
There's quite a bit to talk about when referring to Tsunades ninjutsu capabilities.
Regular Ninjutsu: Even though Tsunade is in her fifties, she looks as if she's in her twenties because she uses a seemingly permanent transformation technique. This technique seems to be an unconscious jutsu seeing as how she doesn't need to use effort to maintain this transformation. For example she still has the technique activated when she's in her sleep. The only time this jutsu is reverted involuntarily is when she runs completely out of chakra.
Medical Ninjutsu: Tsunade is able to flawlessly cure nearly any injury that would be otherwise known as impossible. For example, she's able to heal psychological damage done by Tsukuyomi, and she was able to fully heal Rock Lees spine. She also counteracted the effects of the three coloured pills for Choji which Choza said was impossible. Tsunade was also able to make intense sleeping potions which were colourless so she was the only one who could tell if she was drugging another ninja or not.
Tsunade has also used Medical Ninjutsu to harm others. She uses her knowledge on the human body to direct pin-point punches that could incapacitate someone or leave them internally damaged. Tsunade is also able to create a Chakra Scalpel both for surgery and to inflict internal lacerations and even attack the internal organs.

being her favourite. The slugs can spit acids into smaller slugs, and Tsunade has a mental connection with these slugs, being able to obtain information from afar with just using telepathy. Tsunade can transfer her chakra throughout the slugs and then have the slugs attach themselves to the injured and healing them. This allows Tsunade to heal multiple injured people anywhere at the same time.Taijutsu
Taijutsu is Tsunades main form of attack, and combined with her extensive knowledge of the human body it can pose a deadly threat. An opponent struck by her attacks could suffer damage ranging from broken bones, ruptured organs, or even death. While a medical-nin normally tries to avoid direct battle, Tsunade’s close-range combat skills have been considered unsurpassed, which she uses mainly to intimidate her opponents from approaching her in order to carry out her primary duties as a medical-nin. Tsunade was able to even punch through Madaras Susanoo rib cage and sword. She was able to break through Madaras entire body which forced him to use Wood clones so he wouldn't get sealed.
Since Tsunade applies her medical and psychological intelligence to her fights, she can easily identify an enemies fight patterns and style, coming up with a counteract and taking the advantage seemingly instantly. Usually she tries not to fight so she comes up with strategies that allow her to heal her teammates. On top of this, Tsunade has a considerable amount of chakra at her disposal on top of a very strong life force due to her being a Senju and Uzumaki descendent. For example, even after her muscles were torn, she was still able to move, something Kabuto even said was meritorious. Intelligence
Tsunade is definitely fit for the title of "Hokage" seeing as how she's been shown as a keen thinker and quite observant and analytical during battle. She's able to see through most deceptions and is rarely caught off-guard. 10/10 times Tsunade picks the best suited teams to take on a mission. Her intelligence is most renowned as the best in the world for her expertise in all areas of medicine. She has been credited with creating the official Medical-nin system and the laws that govern them. She can immediately determine what kind of injuries or illness (physical or mental) a person has from a single glance. In addition to this, Tsunade seems to have a vast knowledge of poisons and herbal medicine, as noted by Chiyo who stated that during the Second Great Shinobi War, Tsunade was the only one capable of countering all her poisons perfectly. She also has remarkable knowledge of the human psyche, as she was able to effortlessly undo the damage to Kakashi and Sasuke's minds inflicted by Itachi's Tsukuyomi in a matter of seconds. Tsunade has also proven to be a very capable teacher, having single-handedly trained Sakura and Shizune and passed on much of her knowledge make them become two of Konoha's best medical-nin and field support.
Chakra
Seeing as how Tsunades grandmother was an Uzumaki, it's natural that Tsunade would have a very high reserve of chakra.
Even though Tsunade does have a large amount of chakra, it's not really what I want to focus on, Tsunades chakra control is fantastic. Since a child, she's been able to manipulate her chakra so well to the point where she was able to hold up multiple techniques at once. She can also manipulate a large portion of her chakra into one area to do something like increase the damage given, for example she uses a chakra enhanced taijutsu fighting style, enhancing all of her kicks and punches with chakra on top of her insane Senju strength becoming one of the strongest one punch killers. This is done by building up and releasing her chakra with precise timing upon impact, making it effortless for her to decimate any target.
