Since the sage was an uzumaki so is it possible

Because the Uchihas and Senjus have the Juubi's essence in them, and when Madara combined them (body+ eye of juubi essence) he awakened it, but very very late in his life because it was only residual essence while Rikudou had the Juubi itself inside him.

see...one thing i can say...the sage knew his powers inherited by his elder son could be misused which is why he probably placed an uzumaki seal on that eye(that can explain the swirl pattern on the elder son's eyes). and somehow the juubi's essence was there in the elder son which mixed with the chakra coil of his eyes formed the sharingan(tomoe). This is just a theory though....

then why does naruto have to get kurama's chakra to make the seal like that
the younger son inherited the life-force and yang chakra he could have done it without kurama's help

It is possible that the sage deviced it such that the seal can only be unlocked completely when his physical energy the senju dna gets mixed with it.

The Senjus and Uzumakis cannot be sibling clans for that would contradict Kushina's statement of them being distant relatives, since in Genealogy and the coefficient of holistic relationships, a sibling clan is considered a close relative, no, they did not branch from the Senju Ancestor, the Uzumaki Clan during Mito's era and the Senjus were a cousin clan because they descended not from Rikudou himself, but from Rikudou's Uzumaki relatives. The Uzumaki being the original bloodline and clan of Rikudou Sennin.

Possible, none the less it is a logical impossibility for the Uzumakis to be mete descendants if the Senju Ancestor. Quite the contrary, both the Senju and Uchiha Ancestor descended from an Uzumaki member: Rikudou Sennin
