Are those Edo Nagato and Edo Itachi or not?
If they're Edo and the Senju Bros. aren't, I think Nagato and Itachi could take this - otherwise, Hashirama and Tobirama takes this. By the way, wrong section!

Both Hashirama and Tobirama have fought and defeated the Sharingan, including the Mangekyo. However, neither of them has ever come across the power of the Rinnegan.
Hypothesis: Hashirama and Tobirama will win due to Nagato's immobility and Itachi's outmatched stamina.

As stated above, Itachi is actually the weakest link in this, not in terms of power, but in terms of stamina. Itachi's stamina is ranked 2.5/5 and we've seen manga pages of him in exhaustion in a matter of minutes. Nagato is the only one who can keep up with Hashirama and Tobirama. Also, Itachi is the only non-sensor in this match-up!

In the end, Itachi's poor stamina and the Senju's superior physiology is what gave them the win. Nagato did a great job of pretty much ending the game nigh-tie. READ THE BOTTOM!!!
P.S. In all honesty, I don't believe we've seen enough of Tobirama nor Hashirama to challenge them with Nagato and Itachi. This fight for the Senju was about 45% proof and 55% hype.

Taijutsu Hmmm. Straight on hand-to-hand combat doesn't always happen with Itachi, as his taijutsu is a genjutsu-taijutsu hybrid. However, Hashirama's Senju blood enables enhanced physical strength and much more endurance. Point: Senju.
Genjutsu Itachi has immense Genjutsu prowess, but Hashirama is a sensor and is can disturb his own chakra to break himself out of Genjutsu. Point: Senju.

One thing to note is that Hashirama and Tobirama have fought against countless Uchihas, which means that they probably have means to not get caught in genjutsus or get out of them. While Itachi may have both of them in terms of speed, his stamina isnt near enough to compete with Tobirama and Hashirama.
Also another thing to think about. Teamwork. While Itachi and Nagato were both Akutsuki, beside their short fight together against Naruto and Bee, I dont think they have ever fought with each other. But Hashirama and Tobirama have fought together their entire lives, which combined, they are more than enough to take down Nagato and Itachi.
