Even with Susano'o on,he aimed at himself,Susano'o being transparent he would still watch himself in the mirror.

That and the Amaterasu aimed at his own eyes,destroying them and sayonara!

How about this then, the Amaterasu would spawn ON the mirror, not Sasuke...since technically he is looking at the mirror, not himself. And if the enemy substituted with the mirror, the Amaterasu would spawn right where the enemy used to be (I.e. the mirror)
