Sorry if this has already been said. But maybe Sakura was simply incapable of bearing a child (Problems w/ Uterus?). When Karin was stabbed, her and Sakura shared a moment where they understood each other even though they were on opposite sides. They even cried together. Remembering this, I bet it was Sakura's own suggestion that Sasuke ask Karin to hold her child.
Also they probably didn't have ***. Sasuke may have just given Karin a sample of his own sperm and she mixed it with an egg of her own. This probably all went down in that little DNA lab, and is probably why Karin didn't want Suigetsu poking around in there (she was probably growing Sarada artificially if not in her own womb).
