After thinking about it and re-reading the *********** and narutobase translations, it seems unlikely that Sai and Jugo were actually responding to that comment.
The juubi had just been hardcore hit and everyone's waiting to see the aftermath and standing around in awe of the combined attack. Sasuke took the opportunity to sit back, make a dramatic/cool statement and look evil, which prompted both Sai and Jugo (now that everyone had a second to catch their breaths) to consider Sasuke's earlier statement about wanting to be Hokage.
This isn't to say that Sasuke couldn't be teaming up with Orochimaru still, or that he isn't evil, but I don't think their reactions (or Sakura's) had anything to do with Sasuke's words. If anything, it had more to do with him seeming a little emotionally unstable and reminding them of the time he went homicidal.
