that Orochimaru uses to bestow Juugo's Sage Transformation ability onto his subordinates? We might have an answer.

Above, we see Juugo transferring his chakra into Sasuke, this causes the Zetsu spores Obito had hidden on Sasuke to activate. Implying some correlation between Juugo's cells and Hashirama's. I believe, Orochimaru's enzyme is made up of Hashirama's chakra, as well as Juugo's own.

We've already seen through Sasuke that Juugo's chakra doesn't quite do the trick on it's own.

From here, we know that Sasuke can react to Juugo's chakra, which he now has in greater magnitude, but he's still unable to use that ability. Therefore, Juugo's chakra cannot be the only component. Hashirama's chakra serves to activate Juugo's chakra, as the reverse has already been demonstrated. So, in 661, when Hashirama transferred his chakra to Sasuke, he unwittingly gave Sasuke access to the Sage Transformation ability.

Hmm.... Quite an inherent plausibility. Makes sense given the context. Sasuke already has Juugo's cells when Juugo did the life force transfer technique, but it seemed the magnitude of such was not enough to give Sasuke the ability to absorb nature chakra on his own, but instead, still gave his affinity to Juugo's chakra and Curse Mark.

This might lead to the possibility that the Elder Son's Dojutsu may arise. Given that, if you look at the picture of the Elder Son, we can quite clearly see he has markings around his eyes, almost completely indicative of some kind of Sennin Modo and or Sennin Transformation. That may be the component that one may need to awaken the ESD. As opposed to the Senju and Uchiha's Yang and Yin coming together for the Rinnegan, the overarching amount of Yin from an Uchiha with an Eien No Mangekyou would be stimulated by Nature chakra, and would affect their eyes, bringing them back to how the Elder Sons were.​

I agree. After I had this thought, I instantly found separate meaning in Hashirama transferring "all" of his chakra to Sasuke. As I found that quite strange. Did Hashirama happen to have the exact precise amount of chakra to give Sasuke his mystery Jutsu, or is there some other use? ST would be a decent alternative to Rin'negan, which I've typically been sour on that thought, I came around a bit as I found that it could be a quick way to make Sasuke's fight with Madara a bit more competitive.

I'm not entirely sure on the elder son's doujutsu. I've always felt that it could be the original Mangekyo Sharingan, but it's lack in focus has led me to find it would be somewhat sudden for it to become some greater transformation of the Sharingan. Though Senjutsu used along with the Eternal Mangekyo is definitely intriguing, whether or not it leads to a new and greater doujutsu.
