Obito talked about creating things with shadow style and using light style to invoke life into them. The "biju were but one example..."we don't know of anything else that has/had life that was created by the Sage, so maybe his sons were created with CoAT.
There is also the matter of the misattribution of the source of the Senju and Uzumaki powers, it is a misconception that the Uzumakis share the same traits with the Senjus (life force, chakra) they don't , because sharing implies they come from the same source, they do not. They have similar qualities but their sources are different, the Senju life force and chakra's comes directly from the Younger Son (Juubi's Yang) while the Uzumaki Life Force and Chakra (along with their trademark red hair) comes from their Ohtsutsuki Ancestors.

No, they were not twins. There is a stark distinction between Elder and Younger, and the scarce difference there is from twins being born minutes apart. And they were almost certainly created via Banbatsu Sozo, and not biologically. The allocation of their abilities, split evenly into Yin and Yang, and there relation to the Juubi is way too common and uncanny for it to be of a biological conception.
