Basically anime is short form of "Animation from Japan". It could be based on a manga or not,it doesn't matter.

Actually every animated cartoon made in Japan is called anime whatevery it is based on manga or somthing else some anime are based on a game like school days or even novels and so on.
Also the drawing style is diffrent from normal cartoons so that is also a factor.
Well big eyes are the most notable difference from normal cartoons and other things are also bigger.
