He's part of the reason, but not the whole reason. If you had lived Madara's life, could you honestly tell me that at that point the only thing you'd be worried about was Tobirama?
The strongest Uchiha's always have someone they love dearly, it is through the loss of that love which gives them their power. For Sasuke, it was Itachi. For Itachi, Sasuke. Madara loved his younger brother, he was once one of FIVE siblings. All of them died, even Izuna, whom he had sworn to protect. But Izuna's death seemed necessary for a higher purpose, the clan.
It was Madara's promise to his last and dearest brother that the Uchiha clan would be protected by it's leader. Madara even went as far as to trust his clan's mortal enemies against the will of his little brother, giving up on revenge, giving up on hatred, all for the protection of the Uchiha! Konoha was just an after thought, the clan was his priority!
Tobirama's demeanor posed a danger towards his promise, Madara foresaw the Uchiha's discrimination and possible extermination at his hands, no one but him and Hashirama knew about Tobirama's true feelings. Hashirama was the only one standing against it all, eventually he and Madara would be gone, and that would signal the end for the Uchiha.
Madara took the initiative.
