We all know Oro has already done this before with Yamato and whatever is in that scroll was the ultimate result of his research.

Suigetsu found this in the scroll and quickly thought of taking it to Sasuke though I don't he understands the consequences.

Forgive me if I'm wrong (also feel free to correct), but I don't think that it's been stated explicitly that Senju Hashirama's DNA is needed for the awakening of the EMS->Rinnegan by ET Madara. As I understand it, it's simply the next step in the doujutsu evolution according to the Uchiha tablet that Orochimaru discerned/theorized independently via research with Kabuto going back to when ET Madara was summoned. Thus, the scroll could contain research on the EMS->Rinnegan, but have nothing to do with Hashirama's DNA.

I believe that Hashirama's DNA was simply another component to empower himself that EMS Madara gained independent of the Rinnegan awakening in the past fight at the Valley of the End.
