The scroll and how Orochimaru might have known
We all know that he wanted to master every jutsu in existence. For this, he wanted an Uchiha body with a full Sharingan to reside in. After his failure in getting Itachi's body (and the subsequent trauma of the experience that caused him to start away from Itachi), he set his sights on Sasuke.
It is assumed that after getting Sasuke's body, he would employ the Sharingan to learn the maximum number of jutsus in a span of three years.

Madara, Nagato and the Rinnegan:
When Orochimaru joined the Akatsuki, it is natural that he recognized the Rinnegan Nagato wielded as the legendary eye of the Rikodu Sennin. Kabuto stated that Orochimaru was digging deep into the Uchiha clan secrets and was speculating on the Rinnegan's origin. Eventually, Orochimaru must have deduced the EMS + Hashirama's DNA = Rinnegan equation, which seems to also need some other requirements.

My line of thought should be pretty clear by now.
Orochimaru wanted the Rinnegan and KNEW how it is unlocked. The scroll probably contained the details of this, and of whatever catalysts the process needed.
He intended to take over Sasuke's body after he killed Itachi and was weakened by the fight, expected the fight to open up Sasuke's old wounds-hence giving him the Mangekyou Sharingan and finally wanted to transplant Itachi's eyes to gain the EMS. We know through Tobi's speculations that Orochimaru had access to Hashiramas cells at some point, and hence the Rinnegan.

Suigetsu, like many other shinobi, knows of the Rinnegan only through the takes told about RS. He may know that the faux leader of the Akatsuki possessed it. Hence, his reaction to reading the contents of the scroll were of sheer shock.
If the scroll did contain such information,.the following points would fit:
1) He wanted to give it to Sasuke, and was right that it would agitate the balance of power in the war.
2) He was right to think that Orochimaru crossed the line, 'coz come on bro, it's the Rinnegan, and who knows what Orochimaru would have done with it.
3) Sasuke would ofcourse be very pleased, and Suigetsu was right in thinking that this was a perfect gift.

Ultimately, I believe the scroll is one of the legacies of the Rikodu Sennin, like the Uchiha tablet. If it was simply something written by Orochimaru which Kabuto didn't know and later found out, Kabuto would simply take the scroll along. Hence, Kabuto probably doesn't know of the scroll's existence, but since he worked with the Snake Sannin, he knows of its contents.
That, or the scroll is probably a document and translation of the information on the Uchiha Tablet.
The only question is how Orochimaru connected the dots between the EMS and the Rinnegan. Well here's how imo,
Orochimaru probably said "screw it" after the Akatsuki fiasco, and decided to take Madara's eyes. However, when he found the corpse he noticed the lack of eyes and the fusion of Hashirama's cells. It was obvious that someone beat him to the eyes.
All he had to do was wait and watch for such eyes to show up.
He then must have spotted the tablet left by the Sage, and through what little he knew (there might have been diagrams), he made the connection between the coveted EMS and the Rinnegan he witnessed in the Akatsuki.
