This is a hard one and we don't really know how strong Orochimaru really is even though he was one of the main villains. For now though totsuka blade>Orochimaru and Tsunade.

Of course, Itachi has limits. But they're not exhausted, here.

He's superior to Orochimaru by a good margin, and totally out of Tsunade's reach. Tsunade has two options: Engaging him and trying to distract him with hand-to-hand combat, or staying back and working as a healer.
  • The latter would be useless, since Orochimaru is very well capable of undoing damage by himself. Tsunade wouldn't have much to do as a healer in this fight, and therefore would be totally wasted.
  • The former, however, would be rather useless due to Itachi's predictional abilities or Susanoo and would quickly end with her being entrapped in Itachi's ordinary Genjutsu or Tsukuyomi. Mental damage is unlikely to be healable by Byakugou. In a worst case scenario Itachi will use Amaterasu on her.

That being said, Tsunade isn't much of a threat, here. Orochimaru is neither, as I assume Edo Tensei is restricted in this fight. Amaterasu will prove ineffective on him, thanks to his rebirth-technique, but Tsukuyomi should still show effect in case of eye-contact. Manda and his various huge summonings are incredibly strong, but badly matched against Itachi, considering he can easily control them with his hypnotic Sharingan. Even if he can only control one at a time; taking control of the strongest and making it fight the others or even Orochimaru will mostly render them useless. In case he's pressured by them, Amaterasu will save the day.

That being said, the rest Orochimaru has shown will be blocked by Susanoo or countered with Itachi's extraordinary speed and Bunshin feints. The real danger is Orochimaru's persistance, as it might exhaust Itachi and has a possibility to push him towards his limits. But if he's combining feints with clever use of Genjutsu, which he definitely will, he'll sooner or later create an opening to seal Orochimaru.

After all this you gave no clear assurance that Itachi would defeat Orochimaru. Here mentioned ways in which Orochimaru's attacks would be countered, yet Orochimaru has means to counter Orochimaru's attacks as well. The only issue would be Itachi's blade, in which we don't know the extent of its speed, but will be the deciding factor.

Btw I didn't see Edo being restricted

Orochimaru failed once against Itachis Genjutsu. Paralyzing his opponents with Genjutsus leaves also great openings for counter attacks. Orochimarus offense isn't very great either, Susanoos ribcage or at least the full skeleton should be able to block anything. Since Itachi is aware that Tsunade is a great healer, supporter and can conceivably heal herself due to her medical skills, Amaterasu would be a one shot and Itachi will be aware of that.

If Orochimaru got one shotted once and Tsunade is anything but a threat, Itachi takes this, considering that he will use his Tsukuyomi, Amaterasu and Susanoo here.

Orochimaru wasn't on the defense at all, at that state anyone could be caught by genjutsu. Orochimaru like Kabuto could use his snakes to as his sees to defend against jutsu.

No, Orochimaru was in a normal state, attacked first and failed. Who says that Itachi isn't one shotting him before Orochimaru is really acting? Itachi is known for his feints and illusion he cast in the very beginning of a fight. So who says that Itachi isn't catching Orochimaru and Tsunade in a Genjutsu without they're even noticing it? Just like Deidara who thought to fight Itachi, though it was only an illusion cast by Itachi.

Orochimaru had two chances but did nothing, without Edo Tensei Itachi would beat him with no difficult. With Tsunade, moderate difficult at most since she can nothing do to Itachi, too, since her fighting style is based on Taijutsu and her healing techniques won't be a help here.

Actually an actual attack can't occur under an illusion, Orochimaru was standing at Itachi face-to-face, no attack was shown except for n the anime and that was when Itachi's back was turned. HOWEVER, once it was shown he was in genjutsu, the snake wrapped around Itachi disappeared, meaning the attack never happened really. Nonetheless, Orochimaru was face-to-face with Itachi and for some dumb reason failed to see his genjutsu as a threat. But, that doesn't mean he doesnt have the means to avoid his genjutsu, as shown by Kabuto who used snakes as his eyes to defend against it.

The second encounter Oro emerged drained of chakra basically, handicapped and unaware of the sword Itachi possessed.

In fact Orochiamru has the means to counter any MS techniques easily, the only problem is Itachi's blade as stated.

Did I not? He already fell and will again fall to Genjutsu, that is your assurance. The databook even says that Orochimaru became a small rabit in front of Itachi's superior ocular powers. Using Genjutsu or Karasu Bunshin, like shown against Kabuto who even possessed sensing abilities, is enough to create an opening for Totsuka no Tsurugi. Orochimaru's confession of inferiority is basically enough to end this particular point. Not to mention that it was made at a point where Orochimaru was unaware of Totsuka no Tsurugi being in Itachi's possession.

Don't ridicule yourself by claiming Orochimaru can cover his eyes the way Kabuto did.

And why couldn't he actually?? Btw his eyes weren't covered, he simply used the snakes vision. Even Sauke was aware of this, thus prepping for Oro. I agreed on Itachi's sword being being an issue, tho we don't know of its speed usage.

From what we have seem from Orochimaru (which isn't everything) the abilities he possess presents him as a foe.
