So first off. Your bios:
1. The template has changed slightly a bit along with you being jounin rank now
2. You can mix kgs (such as sharingan/hyuuga etc) with advanced elements (steel, dark, crystal) or Hidden abilities (generally clans line nara, akamichi, hozuki).
3. Specialities are now lit. You can build you bio like a talent tree to specialize in one area to give you buffs.

Advanced Elements can have one Advanced Ninjutsu and one Mode (excluding Sage Mode)
KGs can have one Advanced Ninjutsu and one Mode (excluding EIG)
Mangekyou and higher Doujutsu bios cannot have Sage Mode.
CCs can have one Advanced Ninjutsu OR one Mode. EIG can only be put on clan bios that don't use Natural energy.
Alternatively, if the user does not have a KG, KE, HA, or CC bio, he is able to have two Advanced Ninjutsu on one bio
KG + HA
KG + AE
AE + HA
Due to the nature of the Doujutsu themselves, one cannot pair any Mangekyo or Higher level Sharingan with a HA/AE, nor can Tenseigan.

So basically the specialities allow us to perform techniques faster, with single hand signs or give us a boost of some sort (Defence, Attack, Range etc.) depending on what you choose to be a specialist in. Extra specialities like Fuuin or Medjutsu or Custom fighting stles are not included in the aforementioned list so technically you can have the regular 3 specialisations and whatever else is extra for you.

Say you want to go apex fire, do fire without handseals. You'd have to be primary fire specialist. Same with Genjutsu, you'd have to have a primary or secondary that is genjutsu to get the apex route, following a theme.

My man, you aint just a pretty face it seems. You are correct on both points. S rank is 1500 kumi, though you need you get a general idea of the criteria. You can also do like custom missions, make up your own goals. But first stick to the cannon ones, get a feel for it. Even if you fail, they give you a full run down of why you fail, so you can improve.
