It seems Obito needs to sync his Kamui with a location before being able to teleport there. This must be a new drawback Kishi is introducing. I think Obito will use his left Kamui on Kaguya first, then use his right Kamui to get to Sasuke

On the subject of how Obito knows where teleport to places he hasn't been, my headcanon is that he uses Zetsu to target his teleport, much like how Minato and Tobirama use their seals.
