This is very debatable. But, I am of the opinion that he killed Yura. They merely messed up his mind, but he was physically alive.

That's actually not true, Zetsu can shape himself like anybody and imitate their chakra, but that thing is totally useless in combat. While the transformation used on Yura basically made him Itachi, the only noteable difference was the chakra pool. Compared to most other transformation techniques this one must have heavy drawbacks on the substitute, mainly because it only broke when the imitation was utterly annihilated.

Imprinting another chakra signature into a human, using him for prolonged time as a puppet, forcing the body to physically imitate somebody else (even under prolonged straining combat experience), such heavy changes probably burn out a human being.
