mokuton is genetic,otherwise hiruzen would be able to use all kekkei genkai that require multiple elements.

Wood Style is exclusive to Hashirama, due to him being Ashura's transmigrator and due to his strong chakra that can withstand overwhelming and chakra-taxing kekkei genkai.
Others are just borrowing his powers (Danzo, Yamato and Madara) and look what happened to Danzo, he almost turned into a tree because of Hashirama's dna.
As for Yamato, he's just using a lesser power of Hashi's Mokuton.
Madara is understandable because he's a transmigration of Indra's and that probably enables him to control Hashirama's Mokuton properly.
Besides, they all have one thing in common; they're using Hashirama's DNA.
So wood style is undeniably exclusive to Hashirama.
Mei probably don't have the chakra nor the ability and the proper means (hashi's dna) to gain Mokuton.

Kishi needs to make wood style a kekkei touta (if it isnt) make up a water+earth kekkei genkai. Mud comes to mind but it's not a kkg. Why does she have 2 kkg but not 3? If she only had 1 its understandable but she has 2.

Kishi doesn't make Mokuton as a Kekkei Touta is probably because Yang Release is not really an element. Think of it as a life force from the Juubi itself. Since Hashirama and Madara are both Ashura and Indra's transmigrant, they inherited this power (since Yin-Yang Release is separated into two). Since Mei is not a transmigrant (and inherited Ashura's chakra itself; that what causes Hashirama to be able to use Mokuton. If he isn' a transmigrant, then he will be a fodder), then there's absolutely no way for her to gain Mokuton unless she implanted Hashi's DNA.
For your question about why Mei possess two KKG, well you can safely assume that her father possess one KKG (lava) and her mother possess one (boil release) because it can be passed by bloodlines (e.g Sharingan)

she probably doesn't have the genetic disposition to combine Doton & Suiton. That would be my guess.
