If one who has the Mangekyou in both eyes, but say only uses, the left eye. Will both eyes be effected or just the eye thats being used to cast the jutsu?

That can't be confidently answered. We do know that the eye that is used bleeds and the character ends up covering it in pain, so it could possibly only affect the certain eye, but that's just speculation.

It's hard to say because we don't have enough information on the subject. What I will say though that is there's a strong likelihood that the usage of the double mangekyou accelerates the process of blindness for the user. Obito used spamui freely with his one MS eye (Kakashi too) and they didn't seem to succumb to the same effects of blindness that Itachi and Sasuke (both double MS users) did. Hell, Madara too.

Susanoo in particular is what I believe hastens the process of blindness. Sasuke commented on how it strains every cell in his body and it wasn't until he started using it that his eyesight became an issue.

Agreed with everything except Susano'o.
Susano'o seems to be more of a body technique than an eye technique(granted that the eyes are part of the body)
From what I've observed, it seems that you only need the double mangekyou to unlock Susano'o, not activate it.
What got me thinking this was when Madara pulled the no eyed Susano'o trick.

Well we can say that Kakashi said at 1 point during the war to the 4th that he was going blind in his LEFT eye. But not his right, sure it isnt a Sharingan but its not the 1 casting the jutsu.. Could go as far as whether or not Both sharingan originate from the same user or not

I think your right about the Susano seems to be from the body. In the Mythology or whatever Susanoo was born from the washing of the nose and amaterasu from and eye and tsukyomi from the other. So I'll assume thats what Kishi was going for in his mind.

Probably both are affected. Itachi used Tsukuyomi far more than amaterasu, but both his eyes went blind at the same time.

If you use only 1 eye and get blind then you wake up with rinnesharingan.

Susano'o is doujutsu and you need the eye to release its true potential. Perfect Susano'o.

Now I can't prove that you can activate PS without eyes because it hasn't happened, but being able to activate atleast V2 without eyes should hint at the true nature of the ability. Yes, it's classified as a doujutsu, but it's really just a chakra construct, not necessarily an eye dependant technique, such as Amaterasu or Kamui sniping.

Itachi was going blind and Sauske was going blind by overdoing it so much when he was fighting Kakashi.

So it affects bothe eyes, is the power that takes away the sight

I say it affects both, we've never seen only one eye going blind unless it used izanagi/izanami

Of course Its a physical manifestation of chakra. However, the true power of Uchihas lie in their powerful chakra AND their Eyes. Not either or. So I believe you cannot use Perfect Susano'o without the eyes.

There has never been a MS user using one eye. They both activate at once. Unless you count Tobi hes not in it for obvious reasons.
