he might not have known that killing or witnessing the death of someone important to you could activate the MS and then activated it randomly like when obito used S/T jutsu without him knowing it.

Honestly, it seems to me like he would have unlocked it this way; 1. Let's assume he killed Rin. That's one best friend down.
2. He constantly stated he felt responsible for Obito's death during the gaiden and at other points during the manga. Two best friends.

All in all, it seems that once he learned of the Magnekyo, he probably just trained, thinking the whole time of how his friends died because of his own actions. That, I would think, is how he awakened and learned Kamui.

He could have unlocked at the time of Rin's death, but did not know just like how sasuke was little he unlocked his sharingan at a young age when he chased Itachi at the time of the uchiha massacre.

It must have been plenty of strain on kakashi if he did it by accident, or if he had to kill her because she was betraying the leaf, she was still close to him.
