I think i have one plausible theory now

I think that at one point in time Madara had met Orochimaru. I say this because Madara died within the last 20 years Orochimaru would have been around 30. Who else know more about immortality then Orochimaru. I think that at some point Orochimaru genetically reconstructed a comatose Madara kept alive with a Zetsu completely secret from obito . The rebuilt Madara being the sacrifice for madara's edo tensei. Its never stated how Madara's edo was special just that it was. in the end its still a mystery.

Madara's Edo Tensie is special because old Madara's DNA was scientifically enhanced by Kabuto. The Edo Tensie vessel for Madara was used as body for Madara's soul to come back as young. At the same time, Madara had retained his Rinnegan and all of enhancements as well as knowledge with his prime powers in a young body making him a lot more powerful than he could ever be in his living form.

That's why his ET is special.

1-Madara while coming back alive via Rinne Tensie no jutsu, his Rinnegan disappeared. The reason for that is because when Madara died, he didn't have his Rinnegan because he gave it to Nagato. Rinne Tensie revives you at the same state as you were when alive hence why he lost his Rinnegan. His body did not disappear because Madara's soul took over the Edo Tensie vessel and transformed it into Madara's own body hence why Madara was excited for his blood to move at high speed.

2-Kabuto found Madara's DNA because he along with Orochimaru have stolen a lot of information from the cave allowing them to steal Madara's DNA. That's why Kabuto knows so much about the DNA, Gedo Mazo and Juubi which Madara's hideout contained. Orochimaru was stealing information there and Kabuto absorbed Orochimaru.
