Maybe, but he did receive other wounds than just the one in his chest.

I believe that Madara put Hashirama in his Genjutsu before getting stabbed.
Since no one has yet seen what Madara's EMS can do. I definitely say that Madara's EMS powers is itself Infinite Tsukuyomi which he wants to cast on the Moon through Juubi. Juubi is just a medium to put everyone under his Genjutsu and create a more detailed world by being the Juubi's Jinchuuriki.
In similar way, I think that Madara put Hashirama under his Infinite Tsukuyomi (cast on just an individual) and controlled Hashirama to do what he wants after putting him under the spell. Then he must have obtained the DNA. That is why Hashirama is still confused how someone got Madara's body for his DNA. There must be something that Hashirama did after stabbing Madara. He either must have burried Madara into the valley of end or in the village as a friend. And before he gets out of the spell he died from the effects of his battle with Madara.
It is a simple face that without his full power Madara can't beat Hashirama. I believe Madara never used EMS power before whenever he fought with Hashirama. He was waiting for a moment. And this was the moment since he wanted to obtain his DNA and that's not a simple task.
