Yet we get to know his uncle, Asuma, the third's son.

Are they still alive? Are they Anbu? Are they even Ninja?

Konohamaru is 4 years younger than Naruto, so his parents, at least his mother, were still alive 4 years after the ninetails attack.

Hmm maybe his father died on a mission or something and that is why Konohamaru has a personal teacher (ebisu).
His mom might not be a ninja and that is why we dont see her.
