both know each other capabilities n r prepared for
tsunade with katsuyu slug n kisame with samehada
location: grassland with a small pond nearby

I"m still thinking who would win but..Tsunade is renowned for her monster strength. Her punches are stronger than those of Guy..

Kisame would win, She needs chakra to inflict any damage and to heal herself. Kisame doesn't have to work hard for this fight. We saw when Bee Destroyed Kisame's cheats, and he healed from that. Tsuande wouldnt do that much damage and if she could it would be no problem.

Kisame would steal her chakra and win or make a water dome, fuse with his sword and thats it he wins, no problem

-Kisame versus any ninjutsu/chakra users would usually end up with him win, except for the super powers like, Nagato, Itachi, Madara etc

Tsunade wins. Gai proved that strong taijutsu is kisame's downfall. Tsunade has creation rebirth. She regenerates from all of samehada's attacks, while kisame takes possible killing blows to the face until he passes out.
