I've thought about both of them....they share a personality and both have long hair, but other than that we have no clue....though the fact he wandered the world and only found her hints more towards no familial relationship.
The second one has good hints as well. Just before this we learn about the asauchi who are basically ultimate Zanpaktou, it is also asked who comes first...sword or reaper. Finally it also notes that a sword inherits the personality of a reaper.
Kenpachi cannot communicate with his sword....possibly because it is an extention or his own sword form. His personality would have been created from Unohana's thoughts. This could also explain Unohana's sin: separating the sword with its spirit. The final note on the side is that the mouths on Kenpachi's eye patch resemble the eye mouthed of the asauchi....
Now both instances are entirely theoretical, and a convenient use of information....and honestly I wouldn't really favor either of them. Would especially dislike the second one.
Anyway I'm assuming her sin was to make Zaraki somehow in a state of immortality considering she said every time you cross the road to death (meaning he dies) he gets stronger. Who knows for sure except Kubo!
