In the NUE arc we saw Root being mentioned and how it had a scientific division that was studying Kaguya dimensions with the use of Hashi cells.
Sasuke was also impressed how far Danzou came with the study of Kaguya's dimension.
Sumire's Father was a scientist that was part of this division, primarily for the study and creation of the NUE.
There are some things to notice here.
First, there was a purge of Danzou underlings by Tsunade and the elder council after Danzou was killed off by Sasuke.
Second, the purge was ceased/halted post the war when Kakashi was in power as to not cause political strife when the Shinobi alliance was exhausted. Also not to cause harm for other connected to Danzou like Sai.
Third, the NUE experiment was in use when Danzou was alive and later spearheaded single handily by Sumire's Dad as Root disbanded. There is a whooping 15+ years since then.
Fourth the Kara organization works in groups such as "the elders", the inner, and the outers.
We learnt that Sumire's father acted alone in his NUE research as the other "ROOT" members where elsewhere.
Sai stated that in all likely hood that some Root members might have had spite for Konoha after the disbanding of Root but so far for the last 15 years we haven't seen any retaliation beyond the lone wolf incident of the NUE crisis.
We knew via the Danzou backstory that he had connection and projects not only happening in the LAND of FIRE but also in other areas.

My assumption is this. The elders are the former root members that have some attachment to Danzou. The Inners beyond that one elder are likely experiments or root anbu people that were trained to kill later like the NUE was experimented or created with Kaguya dna or stuff. (Also other things too)
The elders seem to be all about funding the experiments and focused on a set of goals. While the inners seems to have their own plans and are annoyed by the elders.
We saw KK kill one of the elders in chapter 16.
So Kara might've started its beginnings from Root but eventually the things Root created began to take over Kara. Like these numbered inners we see today.
KK could be a Jirayah DNA made individual, akin to Yamato and Hashi.
The Kara girl could be a new experimental dojutsu made by the Root scientific division.
And the Vessal could be the final pinnacle of 15 years of research.
Think of it as how the androids rebelled against Dr. Gero in DBZ.
