Kakuzu wins mid difficulty.
He is superior to Mei in most aspects including stamina, speed, strength, taijutsu, etc.
Mei is pretty versatile and has powerful ninjutsu but Kakuzu can use his attacks at the same time whereas Mei can only do so one at a time. Also the easiest attack Mei has in her arsenal that can hit Kakuzu is her water dragons which are rendered useless by doton domu. Mei's acid mist is rendered useless by either evaporating it with fire or blowing it away with wind. Mei's lava can be cooled down with wind so it will solidify before it hits Kakuzu.
Kakuzu is clearly superior, his masks would outnumber her.
