But what is it that would give him such a huge advantage? Every one of his body enhancing jutsus have some sort of drawback that Kisame could use to take advantage of him.

Its to balance it out because Kakuzu has his hearts and can use his transformation to become a large monster, which will make it much harder for Kisame to defeat alone

I said any ninjutsu that enhances the users body is fair game, meaning that kakuzu's earth grudge fear ninjutsu is allowed so long as he uses it to directly attack kisame or transform.
