I think the best person to answer that is Scorps.

My assumption is that we wouldn't have much information on whether that eye is permanent or not. We don't know how not having a sharingan will affect his performance since he has had it for quite a long time, I'd say almost 8-10 years (don't know the math). I don't know if he would be compromised in any of his abilities. After all he has grown quite adept at using the sharingan that without it, it must be quite a change.

Does anyone know if Minato bio will be allowed to use Sage Mode ?

Rules regarding specific bios will be made as more information is given. Minato will indeed be able to attain his own sage mode, unique to him. Likely somewhat weaker in regards to the normal Sage Mode, but that's left to be determined.

As for Kakashi, that will be decided later.
