In essence, the changes Hagoromo mentioned taking place within individual's who gain or are seeking to gain omnipotence are easily seen. Madara has not only had negative alteration's mentally, that is, the instability and lapses in his rational and behavior currently but there are also anatomical changes. Hagoromo notes that Kaguya suffered identical effect's which brings about the plausibility----sides her absence to face the Jūbi along side her sons. The sin of Kaguya was usurping the fruit and the end result of that or in context 'the consequence she paid' was a rampaging Jūbi.

We have to wait for a little more to be revealed because of the fact that the Sage didn't spoke much about his brother, his mother death and how she died. She technically was still apart of the tree somehow. I know for a fact that the Juubi is derived from the Shinju. Her soul is trap in the tree. Why you think she told Madara to save the tree? I am pretty sure the Juubi was sealed with the Juubi. That may be the problem why it took so long.
