i think he knows what it is.....as he has had it since the begining of the manga...he has been working on developeing the journey to the one piece......(also he planned mostly about the will of D. (source an early interview with him(not done by me of course)

so im pretty sure he and his staff know what one piece is....he just doesnt know what he will have in the way, and how he will reveal it...

Yes. I believe I read somewhere that Oda had the end, and planned the story to be running for 5 years. He just needed to build the path to the end. But it dragged out and extended, and now it's the awesome 16 years run we love (not so much for the time it took, but for the manga itself)
