Madara, Sasuke, and Obito all have one thing in common, they accepted the DNA when on the verge of death.
Think about it, if that weren't the case, then why didn't Kabuto inject it into himself?
Orochimaru could have added it to himself but decided on Yamato and Danzo instead. This leaves me to believe that one of the conditions of accepting the DNA is to be near death. Perhaps that's what Yamato and Danzo experienced.
Too bad no actual explanations were given for their cases.
An alternative is that it might be different for Yamato and Danzo since they are not Uchiha.
If this is the case, then perhaps Danzo and Yamato are closely related to the Senju so they can bypass this near death requirement.
There might be other conditions as well.
