How did Uchiha Itachi gain Mangekyo Sharingan ?

He wiped out his entire clan including his mom and dad. Further Itachi also said that he helped Shisui to accomplish it (Shisui's death). So what more you want to awake the MS.?

U know when Uchiha Shisui died they (the Konoha police force)suspected Uchiha Itachi? But Uchiha Itachi never killed him

here Itachi says uchiha shisui gave him one of his eyes trustiing him 2 protect it, and then shisui died silently to prevent conflicts over his eyes

So if Itachi didnt kill his true friend then how did he activate his mangekyo
cuz to activate a mangekyo u must kill your true/best friend

It was discussed at length before. The Mangekyou Sharingan comes with the death of someone close to you. You don't necessarily have to kill them yourself.

its activated when a family or someone close to you dies...the emotions triggers the sharingan...

SHARINGAN - unlocked from strong emotions (anger)
MANGEKYOU - unlocked from stronger emotions (like death of someone)
ETERNAL MANGEKYOU - unlocked from transplant
RINNEGAN - transplanted eyes + senju DNA???? (maybe, just my theory)
