We don't really know. My guess is that his regeneration ability shortened his life span. No one at that time was able to stand up to Hashirama not even Madara.

He probably played with his wood too much.




But in all honesty I've always felt is was something along the lines of what fluffyguy said above.

it wasnt really said in the manga.. judging by his age after the edo tensei i would say he died from overusing his healing ability and thus shortening his life span or it could be possible he was poisoned by something

He killed himself, that is the only logical explanation. How did Lord second take command? Well after Hashirama defeated and thought that he killed Madara most Uchihas were uneasy about the situation so hashi committed suicide in order to preserve peace.

He lost all of his money and he had an heart attack, afterwards tsunade devoted a life of harnessing his medical ninjutsu, and gambling to earn his money back.

This is plausible but i doubt it. Tsunade's medical ninjutsu is one that seems it would shorten a lifespan and her medical ninjutsu is on a lesser level than hashirama's. on top of that she is way older than hashirama was when he died. with that said i dont think his regeneration was the reason for his death.
