He probably could already tell that Zetsu is comprised of his cells and Madara-sama's will. When Madara-sama stated that Hashirama is not being serious, I believe Hashirama-sama's clone was saying the same thing to Madara given the information he already deducted. Hashirama-sama told Madara to wait! Why is that?!! Because they both knw each others true power and now that Madara seems to e getting serious, this is about to get interesting...in my opinion...
